Q: I want to use this for a 2x8 mattress box that is a loft bed. Will this be strong enough to hold a heavy duty bed?

I believe it is plenty strong enough. I used it to build a twin bed and it is very sturdy even with me on it. Im 6’-3” and over 200 lbs

Q: Does the screw tighten down to secure the rails from coming off

The screw is adjustable. These brackets are awesome.

Q: Does this set of bed bracket come with screws?

Yes. There are two sizes of the screw in the box. I think the longer one is for the flat pieces, and the shorter one is for the angle pieces. Also, you can choose the proper screws for your application.

Q: I have a newer frame that is flat on the ends and a headboard/footboard that require a claw end. Will this work?

I believe these will work.. As long as you habe room to mount these
