Would these work to get rid of fungus gnats?

Be careful how much edible stuff is in there. I recommend springtails as they will eat the eggs. It's not a fast solution.

Wondering if these would work in a Leopard Gecko Bioactive enclosure?

They will! Your gecko might eat them though because they are a larger species. My crestie hasn’t bothered mine, but he also hangs out towards the top of his tank and your gecko would be closer to the ground

What, if any, humidity/heat level to they require?

They are pretty forgiving. Mine range from getting water spill over from my bearded dragon’s “pool” 3x a week. Relationship ve humidity here is very low.
