Q: can it work with jbl eon one?

This unit does not have a battery so requires AC power to operate. The EON 518S also requires an XLR input from a mixer, Only the EON One PRO has a Passthru XLR connector.

Q: What else do I need to buy besides cables? Does it just plugg into my mixer? What cable do I need to buy? Does the mixer need sub out?

Just microphone cables. If you are going to use a pole and high end speaker just above it, then I would suggest buying a short cable, six to twelve feet long. You go straight from your mixer into the sub. There is a built-in crossover that pulls the lower frequencies for the sub to amplify, and you run the short cable from the sub out, to your smaller speakers. I have two for DJing and they sound awesome, even in a gymnasium or outdoors!

Q: Do you have to purchase the speaker pole seperately?

Yes I purchase a pole called onstand because is adjustable for any kind of height

Q: What is the correct polarity for large venues?

Polarity has nothing to do with venue size. Speakers have drivers, which vibrate forward and backward very quickly to make sound. If you have two speakers totally in phase, they will vibrate forward and backward at the same time. If they are totally out of phase, one goes forward while one goes back. This will make the sound quieter than if you had just one speaker. You want your subs, mains, and any other speakers you have to be in phase, if they are not then you can reverse the phase on the subs, which will cause them to move back when they were moving forward and forward when they were moving back, and hopefully that will realign the phase of all of the speakers.
