Is the memory upgradable?

No it can't. DS220J memory is fixed at 512 MB. Current software upgrade requires a minimum of 1 GB according to Synology's own notes. Avoid this model/brand.

what physical size harddrives can be used?

The hard drive "size" is not the physical size of the drive itself but the diameter of the platter inside the drive. A standard desktop HDD is 5.75" x 4" but has a 3.5" platter inside. Hence the name 3.5" drive even though it is physically much bigger. A notebook HDD measures 4" x 2.75" and referred to as a 2.5" drive.

If single drive is installed initially, then 2nd drive is added later. does raid 1 get configured? or does both drive need to be installed together?

I installed one drive initially, and then added a second drive later. The second drive was automatically added as a Raid 1 drive.

Can it be used as a datastore for esxi or backup VMs?

Technically you can but not recommended. You will likely get terrible performance because the CPU and memory is not designed for this use case. 512MB RAM is very low for a datastore. This model is really geared towards traditional file sharing and media streaming. You want something with higher CPU and read/wrtie caching like the DS218/DS218+.
