Will it fit a jeep gladator with a truck bed tent

It fits a Toyota Tacoma with an AT Overlander Atlas topper. The problem may be snugging the two tents together if you want both open for access. It will snug nicely over a topper door which you won't have and up to the bottom and sides of the tailgate without issue.

I see it says jeep hard tops, but will it also work for soft tops?

will this work with a Thule cargo roof box? I noticed suction cups in the video.

It does have suction cups but the roof box should not interfere. In my opinion. You have a lot of leeway for placement.

When you drive away does the tailgate opening zip closed?

Yes it fully zips closed as a second door. Love this tent BTW!!
