Q: After you applied this product to a pipe thread and placed the nut, do you have to wait, or can you start running the water right away?

Depends on how much pressure there is. I know if you put it on a airhose fitting and then pressurize it right away, it won't hold and start to push it out of the threads. I think it takes 24hrs to setup fully.

Q: Use on drinking water systems?

Not sure, I used it on air lines outside. I would use Teflon tape on water.

You would use one or the other, you wouldn't use both... this would take the place of teflon tape.

Q: Can this be used on a high pressured air mattress hose? I need something to seal the threads, but not close off the intake/outtake capacity of hose.

Yes, you should be able to use it on air mattress hose, as long as they're threaded and not just push-on type connections
