Yes it should work, but depending on the motherboard BIOS it may not be supported as a boot device.

The 2013 Mac Pro doesn’t use a standard PCIe anyways so this wouldn’t serve much purpose.

Yes, the motherboard needs to support booting from NVMe. Most motherboards from Intel after 2013(so 2014+) support it, some other ones have support that are older(I run an Intel Core i5-4750t on an ASRock motherboard and it had support for NVMe added in December 2017) You'll probably need to flash any motherboard prior to 2015 for support, though(Assuming the mobo has any updates that support NVMe booting)
