Is this bagpack good for packing daycare stuff for a toddler

It depends on what you want to pack. I used it as a "diaper" bag when my daughter was about 1.5-2. I easily fit several disposable diapers, some wipes, a sippy cup, a snack, and a change of clothes in it

Check Chubbiee bag tags on amazon. They are of good quality with very cute patterns. I use them for everything my kids bring to kindergarten and preschool.

Does it fit a plastic folder?

Not really. It would bend at the corners.

Will a 12 inch insulated straw cup fit in the cup holder pouch?

You mean un the side pocket? I guess it depends on the diameter but it's probably too tall and will fall down. I stuck in small drinking glasses.
